ABSTRACT

Objective: To detect the expressions of proto-oncogene protein FOSB and cyclin protein P16 in the different molecular subtypes of breast invasive ductal cancer and their correlation. Methods: The tumor tissues of 138 patients diagnosed with breast invasive ductal cancer by postoperative pathological examinations in Zhongshan Bo'ai Hospital from August 2013 to March 2019 were collected with normal breast tissues as control. Immunohistochemical staining was performed for the expressions of FOSB and P16 to investigate their correlation in the breast cancer. Besides, their expressions and their correlations in the different molecular subtypes of breast cancers, i.e. Luminal A, Luminal B [According to the expression of human epidermal growth factor 2 (HER2), it can be divided into HER2- and HER2+.], HER2 positive subtype, and triple-negative breast cancer (TNBC), were also detected. Results: ① The expression level of FOSB in 138 cases of breast invasive ductal cancer tissues was negatively correlated with P16 (r= -0.181, P=0.033). ② The expression of FOSB was significantly higher in Luminal A than those in other subtypes (P=0.028, P=0.033, P=0.001, P=0.010), while the expression of P16 was significantly higher in TBNC than those in other subtypes (P=0.025, P=0.005, P=0.008, P=0.011). ③ The expression level of FOSB in TBNC was negatively correlated with the expression of P16 (r=-0.566, P=0.018). Conclusion: The expressions of FOSB and P16 in the different molecular subtypes of breast invasive ductal cancer are different. The expression level of FOSB significantly increases in Luminal A, and that of P16 significantly increases in TBNC. There is a negative correlation between the expression levels of FOSB and P16 in TBNC.

ABSTRACT

@#【Objective】To investigate the expression of FosB in invasive breast cancer and its correlation to the ultrasound findings and clinical significance.【Methods】Immunohistochemical and ultrasound examination were performed for 155 cases with invasive breast cancer,36 cases with paracancerous tissue and 30 cases with normal breast tissue. To investigate the correlations between the different expression of FosB with histological grade ,lymph nodes,and ultrasound findings.【Results】①The expression of FosB in paracancerous tissue and normal breast tissue was higher than that in invasive breast cancer(P<0.01). ②Based on immunohistochemical staining,high positive expression rate of FosB was associated with low histological grade,ER positive and PR positive(P<0.05). There was no correlation between FosB and age,tumor diameter,lymph nodes,Her2 and clinical stages(P>0.05)③The expression of FosB was higher in microcalcification positive and lymph nodes bigger tissues(P<0.05). There was no correlation between FosB and tumor size, regular shape,boundary,posterior attenuation ratio and blood flow(P>0.05).【Conclusion】The expression of FosB in invasive breast cancer was lower than that in paracancerous tissue and normal breast tissue. The different expression of FosB was correlated with histological grade,ER positive and PR positive. To study the expression of FosB in invasive breast cancer may be helpful for differential diagnosis and targeted treatment. The different expression of FosB was correlated with microcalcification,lymph nodes,and combined FosB with ultrasound findings may contribute to prognostic evaluation of breast cancer.

ABSTRACT

OBJECTIVE@#To evaluate rectal toxicity of radiotherapy for prostate cancer using a novel predictive model based on multi-modality and multi-classifier fusion.@*METHODS@#We retrospectively collected the clinical data from 44 prostate cancer patients receiving external beam radiation (EBRT), including the treatment data, clinical parameters, planning CT data and the treatment plans. The clinical parameter features and dosimetric features were extracted as two different modality features, and a subset of features was selected to train the 5 base classifiers (SVM, Decision Tree, K-nearest-neighbor, Random forests and XGBoost). To establish the multi-modality and multi-classifier fusion model, a multi-criteria decision-making based weight assignment algorithm was used to assign weights for each base classifier under the same modality. A repeat 5-fold cross-validation and the 4 indexes including the area under ROC curve (AUC), accuracy, sensitivity and specificity were used to evaluate the proposed model. In addition, the proposed model was compared quantitatively with different feature selection methods, different weight allocation algorithms, the model based on single mode single classifier, and two integrated models using other fusion methods.@*RESULTS@#Repeated (5 times) 5-fold cross validation of the proposed model showed an accuracy of 0.78 for distinguishing toxicity from non-toxicity with an AUC of 0.83, a specificity of 0.79 and a sensitivity of 0.76.@*CONCLUSIONS@#Compared with the models based on a single mode or a single classifier and other fusion models, the proposed model can more accurately predict rectal toxicity of radiotherapy for prostate cancer.

ABSTRACT

Objective To explore the CT dominant phase and optimal classification model in differenting clear cell renal cell carcinoma (ccRCC) from fat‐poor angiomyolipoma (fpAML) through quantitative multiple‐phase CT radiomic features analysis. Methods Clinical and imaging data of 195 cases pathologically confirmed ccRCC (n=131) and fpAML (n=64) were retrospectively studied. All the patients underwent non‐contrast enhanced CT scans and dynamic multi‐phase (corticomedullary phase, medullary phase and excretion phase) contrast‐enhanced CT scans. Regions of interest (ROIs) were manually delineated based on the selected image slices with the maximal diameter of the lesion using ITK‐SNAP software, followed by the acquisition of candidate CT radiomic feature sets from each phase with statistically significant differences by using Mann‐Whitney U test. Then, using the synthetic minority oversampling technique (SMOTE), 232 classification models which are composed of 29 different feature selection algorithms (top 10 features were chosen by the backward elimination method) and 8 different classifiers were constructed. Employing the 5‐fold cross‐validation method, the performance of each classification models for each phase was evaluated using accuracy (ACC), sensitivity (SEN), specificity (SPE) and area under receiver operating characteristic curve (AUC), to acquire dominant CT phases and the optimal classification models for distingushing ccRCC and fpAML, along with the key imaging radiomic features. Results In this study, the mean maximal diameter of ccRCC and fpAML lesions were (3.9±1.4) cm, and (3.5±1.7) cm, respectively, and there was no statistically significant difference in the size of the tumor between two groups (P>0.05). From 102 initial imaging feature sets, the total number of candidate imaging feature sets (P<0.05) were:non‐enhanced phase (n=26), corticomedullary phase (n=71), medullary phase (n=68), excretion phase (n=62). Among the 232 classification models through different combination of classifiers and feature selectors, the amount of classification models which achieved the maximum of AUC value (AUCmax) from different CT phases were: non‐enhanced phase (n=106, 45.7%), corticomedullary phase (n=94, 40.5%), medullary phase (n=23, 9.9%), excretion phase (n=9, 3.9%). Imaging features from non‐enhanced phase and corticomedullary phase yielded higher performance compared with medullary phase and excretion phase, with the corresponding optimal prediction models were SVM‐fisher_score (AUC: 0.897, ACC: 83%, SEN: 84%, SPE:80%) and Logistic Regression‐RFS (AUC: 0.891, ACC: 83%, SEN: 81%, SPE: 89%), respectively. Conclusions The quantitative imaging features from non‐enhanced and corticomedullary phase have better performance among proposed classification models than that from medullary phase and excretion phase. Furthermore, it is feasible to acquire proper combination of feature selection and classifiers to achieve high performance in identifying ccRCC and fpAML.

ABSTRACT

Objective To analyze the clinical features of invasive Klebsiella pneumoniae liver abscess syndrome (IKLAS). Methods The clinical data of IKLAS patients diagnosed in China-Japan Friendship hospital from January 2010 to October 2017 was analyzed retrospectively. Results There were 9 patients with IKLAS including 6 males and 3 females aged (56. 77 ± 10. 28)years old. Four cases with diabetes mellitus,one case with impaired glucose tolerance,and two cases with cholelithiasis history. Single abscess (7 / 9),multiple abscess(2 / 9). Involved organs:lung(7 / 9),eyes(3 / 9),soft tissue (1 / 9),joint(1 / 9)and nerve system(1 / 9). Laboratory test showed an elevation of the white blood cell count (8 / 9),the al-anine aminotransferase (9 / 9)and the PCT (7 / 9 ),and only one ESBL positive Klebsiella pneumonia. Three cases with en-dophthalmitis lost their sight,and one with meningitis died. Conclusion IKLAS is common in people with diabetes,and it may be associated with multiple sites of metastasis of the body and lungs are the most involved organ.

ABSTRACT

Objective To assess the clinical efficacy and safety of transurethral plasmakinetic enucleation of prostate (PKEP) and suprapubic transvesical prostatectomy (SPP) in the treatment of large volume benign prostatic hyperplasia (BPH).Methods A total of eighty patients with large volume BPH were selected in the First People''s Hospital of Xuzhou from March 2014 to December 2016,and they were randomly divided into two groups,the PKEP group and SPP group,with 40 cases in each group.The comparison was made between the two groups in terms of operation indicators,postoperative complications followed up for 6 months after surgery and other related curative effect indexes.Results In the SPP group,the intraoperative blood loss was significantly higher than that of the PKEP group ((215.7±10.4) ml vs.(75.6±9.2) ml,t=6.541,P0.05).But the catheterization time after operation,bladder irrigation time and length of hospital stay were significantly longer in the PKEP group than in the SPP group ((6.3±1.8) d vs.(2.4±0.6) d,(5.5±1.4) d vs.(1.3±0.6) d,(7.7±2.3) d vs.(3.1±0.7) d,t=4.357,2.542,2.975,P0.05).The rate of complication occurrence in the PKEP group were lower than that of the SPP group (P<0.05).Conclusion PKEP was effective in the treatment of BPH,with less bleeding loss,fewer complications and quick recovery.It can significantly improve the life quality of the patients and will be an ideal treatment for large volume BPH.

ABSTRACT

Objective To explore the effect of group mind-games and group counseling on training burnout among soldiers who quickly marched to the plateau for the first time .Methods Totally 399 soldiers who quickly marched to the plateau for the first time were divided randomly into to the control group ( n=201 ) and test group ( n=198 ) .The test group had particinated in group mind-games and psychological counseling twice a week for a total of 5 weeks, while the control group received no counseling .Soldier training fatigue questionnairs were used to compare the difference between the two groups before and after group mind-games and psychological counseling .Results ①The total scores of training burnout and the scores of all the factors of soldiers before counseling in the two groups was of no statistical significance (P>0.05). After counseling , the total scores of training burnout and the scores of all factors in test group were remarkably lower than those in control group,and the difference was of statistical significance (P<0.05).The total scores of training burnout and the scores of physical and psychologica exhaustion and alienaties decreased significantly after training in test group ( P<0.05), but the change was of no statistical significance in control group (P>0.05).②Soldiers who had served 1 to 2 years or over 9 years had significant difference in the scores of training burnout , physical and psychologica exhaustion ( P<0.05).Soldiers who had served 3 to 8 years had significant difference in the scores of training and the scores of each factor after counseling(P<0.05).Conclusion Group mind-games and psychological counseling can effectively help alleviate the training burnout of soldiers who quickly march to the plateau for the first time.

Objective Transcription factor forkhead box L 2 (FOXL2) is a key regulator of granulosa cells (GCs) estrogen syn-thesis and function maintenance .However, the FOXL2 protein expres-sion and function regulation mechanism are unknown .We explored how DNAJB11 regulates estrogen synthesis of granulosa cells with immunoprecipitation , immunofluorescent staining and luciferase re-porter gene. Methods The expression and localization of DNAJB 11 was detected by immunohistochemistry staining in isolated mouse ovary tissues .we use immunoprecipitation , immunofluorescence staining and luciferase reporter gene assay to investigate the mechanism of DNAJB11, a member of the endoplasmic reticulum Hsp 40 /DnaJ family, regulating the estrogen synthesis in granulosa cells . Results DNAJB11 is expressed in the mouse ovary and granulosa cells .Follicle-stimulating hormone (FSH) promotes DNAJB11 ex-pression in a time and concentration dependent manner and induces endogenous DNAJB 11 protein translocation from the ER to the nu-cleus in KGN cells.Moreover, Adenovirus-mediated overexpression of DNAJB11 did not affect the proliferation of granulosa cells .How-ever, the concentration of estrogen in granulosa cells was affected by concentration -dependent and subcellular localization-dependent manner (10749±801.7 pg/mL vs 14217±1218.0 pg/mL P<0.01).Immunoprecipitation assay confirmed that DNAJB11 binds to FOXL2 in granulosa cells .When overexpressed in the nucleus of granulosa cells , DNAJB11 could significantly enhance the stability of FOXL2 (P<0.05) and promote FOXL2-mediated activity of Cyp19A1 promoter (P<0.01), while the expression of DNAJB11 in the nucleus increased the expression of Cyp 19A1 protein by 1.5 times ( P<0.05) . Conclusion These results demonstrate that DNAJB 11 was a new binding molecule of transcription factor FOXL 2 and regulated FOXL 2 protein stability and transcription activity .

ABSTRACT

<p><b>OBJECTIVE</b>To propose arectal toxicity prediction method based on deformable surface dose accumulation.</p><p><b>METHODS</b>The clinical data were collected retrospectively from 42patients receiving radiotherapy for cervical cancer. With the first fraction as the reference, the other fractions of rectum surface were registered to the reference fraction to obtain the deformation vector fields (DVFs), which were used to deform and sum the fractional rectal doses to yield the cumulative rectal dose. The cumulative rectal dose was flattened via 3D-2D mapping to generate a 2D rectum surface dose map. Two dosimetric features, namely DVPs and DGPs were extracted. Logistic regression embedded with sequential forward feature selection was used as the prediction model. The predictive performance was evaluated in terms of the accuracy, sensitivity, specificity, and the area under the receiver operating characteristic (ROC) curve (AUC).</p><p><b>RESULTS</b>Significant improvements for rectum surface DIR were achieved. The best predictive results were achieved by using both DVPs and DGPs as the features with a sensitivity of 79.5%, a specificity of 81.3% and an AUC of 0.88.</p><p><b>CONCLUSION</b>The proposed method is feasible for predicting clinical rectal toxicity in patients undergoing radiotherapy for cervical cancer.</p>

ABSTRACT

OBJECTIVE: To investigate the relationship among perceived stress,sleep and fatigue status; and to analyze the effect of sleep status in mediating and regulating perceived stress and fatigue status in soldiers that were acutely exposed to high altitude for the first time. METHODS: A total of 700 soldiers that were acutely exposed to high altitude for the first time were investigated with convenient sampling. They were evaluated using Perceived Stress Scale,Sleep Status Self-evaluation Test,and Fatigue Scale-14. The mediating effect was analyzed by structural equation model,and the moderating effect was analyzed by hierarchical regression analysis. RESULTS: The total score of fatigue among soldiers that often took part in recreational activities was significant lower than that who did not( P < 0. 01). The total score of fatigue among soldiers that had training injury was significant higher than that who had not( P < 0. 01). The total score of fatigue among soldiers that had high training expectations was significant higher than that that didn't have high training expectations( P < 0. 01). Sleep status,sense of overload and training expectations had remarkable predicting effect on fatigue status among soldiers that were acutely exposed to high altitude for the first time( P < 0. 01). Sleeping status has a complete mediating effect on the sense of prediction,sense of overload and fatigue status( P < 0. 01),and has a moderating effect on the sense of control,sense of overload and fatigue status( P < 0. 01). CONCLUSION:s Perceived stress and sleeping status had close relationship with fatigue status among soldiers that were acute exposed to high altitude for the first time. Sleeping status has intermediary and regulating effect between senses of overload and fatigue state.

ABSTRACT

Objective To study a novel method for the high-dose-rate brachytherapy (HDR) CT image to the intensity modulated radiation therapy (IMRT) CT image deformable image registration and dose accumulation.Methods The applicator in the HDR CT image is first segmented and removed,then a deflation step is performed on the applicator-free HDR CT image by solving the Navier-Stokes equation.Demons algorithm is utilized to register the deflated HDR CT image to the IMRT CT image,along with the HDR dose.The deformed HDR dose is then added on the IMRT dose and yield the final accumulated dose.Results The HDR CT image and IMRT CT image,as well as the corresponding dose distribution,from five cervical cancer patients are used for evaluation of the proposed algorithm,the results show that the proposed method can effectively get rid of the influence of the applicator and produce an accurate accumulated dose.Conclusions Dose accumulation and supervision is an important step in adaptive radiotherapy for accurate dose delivery and treatment plan re-optimization.The proposed method in this study can effectively accumulate the HDR dose to the IMRT dose domain,and the accuracy is proved to be sufficient for clinical needs.

ABSTRACT

<p><b>OBJECTIVE</b>To study the immune repair effect of umbilical cord mesenchymal stem cells (UC-MSC) on inflammatory disorders and thrombophilia state of MRL/lpr mice by detecting the expression change of peripheral blood CD4(+) CD25(+) T cells and the levels of plasma inflammatory cytokines TNF-α, IL-6 and thrombosis indicators TF, FIB.</p><p><b>METHODS</b>Twenty five MRL/lpr mice were divided into control (C) group, UC-MSC one time treatment (UT1) group and UC-MSC three time treatments (UT3) group. UC-MSC cell suspension was injecled via tail vein and these mice were feeded in SPF environment. The blood samples were taken from the mice every 2 weeks after 16(th) week. FCM was used to detect the expression of CD4(+) CD25(+) T cells in peripheral blood, ELISA assay was used to detect the levels of inflammatory cytokines TNF-α, IL-6 and thrombosis indicators TF, FIB.</p><p><b>RESULTS</b>The expression of peripheral blood CD4(+) CD25(+) T cells in treatment groups increased during 16(th) to 18(th) week, dropped and tended to be stable since 20(th) week, and lower than those in control group. The levels of plasma TNF-α and IL-6 in treatment group decreased since 16(th) week and significantly lower than those in control group (P < 0.05). The levels of plasma TF and FIB in treatment group decreased since 16(th) week. The level of plasma TF in treatment group was significantly lower than those in control group (P < 0.05) since 18(th) week.</p><p><b>CONCLUSION</b>UC-MSC can repair the immune inflammatory microenvironment disorders of MRL/lpr mice through its immunomodulatory effect. UC-MSC contribute to repair of immune inflammatory thrombophilia of MRL/lpr mice.</p>

ABSTRACT

Objective The CYP19 gene product enzyme aromatase mediates the conversion of the androgen testosterone to es -tradiol.The aim of this study is to investigate whether the CYP19A1 gene single nucleotide polymorphism (SNP) is associated with the susceptibility to polycystic ovary syndrome (PCOS) and serum hormone levels. Methods We conducted a case-controlled study, which included 373 PCOS patients and 313 healthy controls.We genotyped SNP rs2899470 in the subjects using the polymerase chain re-action-restriction fragment length polymorphism ( PCR-RFLP) method and analyzed the frequencies of genotypes and alleles as well as the association of different genotypes with age , menarchal age, body mass index (BMI), and serum levels of hormones. Results The gen-otypic distributions of rs2899470 GG, TG, and TT in the PCOS women were 44.5%, 49.6%, and 5.9%, respectively, significantly dif-ferent from 39.3%, 48.6%, and 12.1%in the healthy controls (P=0.013).The frequency of the G allele was 69.3%in the former, remarkably higher than 63.6%in the latter (P=0.025).The rs2899470 genotypic frequencies were associated with the serum E 2/T lev-els in the PCOS patients. Conclusion SNP rs2899470 in the CYP19A1 gene is associated with the susceptibility to PCOS , and so is the genotype of rs2899470 with serum E2/T levels, which may be attributed mainly to the reduced activity of aromatase .

ABSTRACT

This study was purposed to observe the influence of umbilical cord mesenchymal stem cells (UC-MSC) on the peripheral blood CD4(+)CD25(+)regulatory T cells (Treg), Th17 cells and neutrophils in rats with collagen type II-induced arthritis(CIA), and to explore the regulating effect of UC-MSC transplantation on immunocyte subgroup. The rats wee divided into 3 groups: CIA group (model group), UC-MSC treated group and blank control group. The CIA rats were injected with UC-MSC via tail vein. The percentage of CD4(+)CD25(+) cells in peripheral blood and the expression of NCD11b on neutrophil surface in CIA rates was detected by flow cytometry (FCM), and the serum interleukin-17 (IL-17) was observed by enzyme-linked immunosorbent assay (ELISA). The results showed that the mean fluorescence intensity(MFI) of NCD11b and the level of IL-17 in the model group were significantly higher than those in the blank control group, and the ratio of CD4(+)CD25(+) cells were significantly lower (P < 0.05). The MIF of NCD11b and the level of IL-17 in the UC-MSC treated group were significantly lower than that in the model group (P < 0.05), while the proportion of CD4(+)CD25(+) Treg increased (P < 0.05). Since the fifth week, the above indicators in the UC-MSC group have almostly approached the control group. It is concluded that the UC-MSC can increase peripheral blood Treg proportion in CIA rat, inhibit the secretion of Th17 and the activity of neutrophils, reduce the immune inflammation reaction, decrease the release of proinflammatory factor, and induce immune reconstruction.

ABSTRACT

<p><b>OBJECTIVE</b>We propose a new iterative reconstruction method based on split-Bregman method with tight frame regularization for effective and accurate reconstruction of the sparse-view cone beam CT image.</p><p><b>METHODS</b>A tight frame was chosen as the regularization term for the objective function, so that the image reconstruction involves only the minimization of an objective function according to the compressed sensing theory. We utilized the split-Bregman method to tackle the task of minimization in three steps: (1) a fast calculation of the forward projection matrix; (2) introducing an intermediate variable to transform the non-differentiated L1 regularization term into the differentiated L2 regularization problem, and solving the target function using conjugate-gradient method; (3) updating the intermediate variable using shrinkage formula from Bregman method.</p><p><b>RESULTS</b>Digital and physical phantom experimental results suggested that our new approach had great advantages in terms of image quality, reconstruction time, and applicability.</p><p><b>CONCLUSION</b>The proposed method can accurately reconstruct CBCT image with limited data to lower the X-ray dose and accelerate the calculation speed in comparison with the POCS method.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the status of HLA-DPA1 and DPB1 matching for unrelated donor-recipient pairs matched at high-resolution allele level for HLA-A, B, C, DRB1 and DQB1 loci.</p><p><b>METHODS</b>A total of 76 unrelated donor-recipient pairs matching at allele level for HLA-A, B, C, DRB1 and DQB1 loci were subjected to HLA-DPA1 and DPB1 sequence-based typing (SBT). HLA-DPA1and DPB1 matching status at high-resolution allelic level was also analyzed.</p><p><b>RESULTS</b>The allelic identity ratio for single HLA-DPA1 and DPB1 were 17.1% and 9.2%, respectively. HLA-DPA1 and DPB1 allelic identity ratio were both very low. The majority of unrelated donor-recipient pairs (73.7%) had an incompatibility at 1 HLA-DPA1 allele, 9.2% of pairs had an incompatibility at 2 DPA1 alleles. As for the high-polymorphic HLA-DPB1 gene, 57.9% of studied donor-recipient pairs had an incompatibility at 1 HLA-DPB1 allele, almost 1/3 (32.9%) of them were completely incompatible. When HLA-DPA1 and DPB1 genes were analyzed together, the donor-recipient pairs matched at 2/4 was the most common (51.4%), 4/4 allelic complete matched pairs accounted for 5.6%, and 0/4 matched pairs accounted for 8.3%.</p><p><b>CONCLUSION</b>Our results indicated that the ratio of HLA-DPA1 and DPB1 complete match in the unrelated donor-recipient pairs matching at allelic level for HLA-A, B, C, DRB1 and DQB1 loci were very low. The effect of HLA-DPA1 and DPB1 matching status on clinical unrelated stem cell transplantation still needs to be elucidated.</p>

ABSTRACT

An algebraic image reconstruction from few views using bilateral-filtering iterative method was proposed due to the problem of computed tomography insufficient data in the present study. In each iteration reconstruction, we first used algebraic reconstruction technique (ART) algorithm to reconstruct an image, ensuring the non-negativity of the reconstructed image at the same time, and then performed bilateral-filtering to the above-mentioned image. In order to improve reconstructed image quality and accelerate the convergence speed, we developed a modified bilateral-filtering method. Shepp-Logan simulation experiments and real CT projection data reconstructions showed the feasibility of the algorithm. The results showed that, compared with the traditional methods of filtered back projection (FBP), ART and GF-ART,the proposed method has a higher signal-to-noise ratio, and maintains more effectively the image edge information.

ABSTRACT

microRNAs (miRNAs) are a family of important small non-coding RNA molecules, which participate in the post transcriptional gene regulation. In this review, the numbers and chromosomal distribution of chicken miRNAs, and the regulation and function of chicken miRNAs in immune, embryo development and virus infection were reviewed. Additionally, the applications of chicken miRNAs were also discussed briefly. We hope it can provide references for further study and use of miRNAs in poultry husbandry fields.
